ABSTRACT

The increasing demand of Chinese materia medica could not be supplied by wild resource, and the cultivated medicinal materials become popular, which led to decreased quality of many medicinal materials due to the difference of the circumstance between the wild and the cultivated. How to improve quality becomes key points of Chinese medicine resource. The leaves of Scutellaria baicalensis were sprayed with H₂O₂, the activities of superoxide dismutase (SOD) and catalase (CAT) changed little, but there had been a marked decrease of peroxidase (POD) and ascorbic oxidase (APX), which showed that the antioxidase system declined. Meanwhile, H₂O₂, as enhanced the expression of phenylalnine ammonialyase (PAL) and β-glucuronidase (GUS) as well as activity of PAL, promoted the biosynthesis and biotransformation of flavonoids. At the day 2 after treated, H₂O₂ of 0.004 μmol·L⁻¹ the contents of the baicalin and the wogonoside decreased slightly, but the contents of the baicalein and the wogonin increased significantly, the baicalein from 0.094% to 0.324%, the wogonin from 0.060% to 0.110%, i. e. increased 246% and 83.3%, respectively.

ABSTRACT

With Bupleurum smithii var. parvifolium and B. scorzonerifolium as test objects, in order to provide a theoretical basis for the introduction and domestication of B. smithii var. parvifolium, the growth and development dynamics of seedlings, biomass accumulation, the content of malonaldehyde(MDA), the activity of antioxidase such as SOD, POD, CAT and APX between them were comparatively analyzed by direct sowing culture in the open field. The results indicated that the morphological index and the biomass accumulation of B. smithii var. parvifolium such as root diameter, root length, plant height and leaf number were inferior to B. scorzonerifolium, the antioxidase SOD and POD activity of B. smithii var. parvifolium was significantly inferior to B. scorzonerifolium (<0.05), the antioxidase CAT and APX activity of B. smithii var. parvifolium was inferior to B. scorzonerifolium but the difference wasn't significant, while MDA content was superior to B. scorzonerifolium(<0.05). Thus, compared with cultivated B. scorzoneri folium, the plant growth velocity of wild B. smithii var. parvifolium was relatively slower and its resistance was relatively weaker after introduction and domestication.

ABSTRACT

Objective: In order to explore the anti-oxidant capacity of wild and cultivated ginseng, the anti-oxidase activities and antioxidants contents on ascorbic acid-glutathione (AsA-GSH) cycle were compared. Methods: The activity of superoxide dismutase (SOD) was detected by NBT method and the activity of catalase (CAT) was detected by potassium permanganate titration. The anti-oxidase activities and anti-oxidants contents on AsA-GSH cycle were tested using spectrophotometric determination. The contents of glutamate (Glu), cysteine (Gly), and glycine (Cys) were tested by automatic amino acid analysis. Results: The activities of SOD and CAT in wild ginseng were higher than that of cultivated ginseng. In AsA-GSH cycle, the activities of anti-oxidase such as ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), and glutathione reductase (GR). And the contents of antioxidants such as glutathione (GSH), ascorbic acid (AsA), and dehydroascorbic acid (DHA) were also more in wild ginseng. And then the contents of amino acids (Glu, Gly, and Cys), the precursor of GSH synthesis were high expression in wild ginseng to keep the balance of AsA and GSH recirculation. Conclusion: The anti-oxidase activities and antioxidants content on AsA-GSH cycle in wild ginseng are higher than those in cultivated ginseng, which may be leaded to the antioxidant capacity of wild ginseng is stronger. It will provide a theoretical basis for efficacy differences research on wild and cultivated ginseng.

ABSTRACT

Aim To investigate the protective effect of riboflavin on ischemia brain damage and the mecha-nism.Methods The in vivo experiments were pro-cessed in male SD rats .Rats were randomly arranged into control group , model group and riboflavin group . The rats in riboflavin group were intraperitoneally in-jected riboflavin at the dose of 1 mg? kg -1 for seven consecutive days .Then the rats in model and riboflavin groups were carried out middle cerebral artery occlu-sion( MCAO) operation.After 24 h, all rats were sacri-ficed and the brain tissue was dissected to observe the infarct area, the edema and the ultrastructure damage . The brain tissue was dyed by triphenyl tetrazolium chloride .The brain edema was observed by the weight of ischemia-side semi-brain.The ultrastructure was ob-served by electron microscope .The in vitro experiments were processed in primary culture neurons by exposed to oxygen and glycose deprivation ( OGD) .The viability of neurons was assayed by MTT method .The enzyme activity of superoxide dismutase ( SOD) , catalase ( CAT)and glutathione peroxidase ( GSH-Px ) was assayed to explore the mechanism .Results Riboflavin signifi-cantly decreased the infarct area ( P<0.01 ) , inhibited the brain edema ( P <0.01 ) and inhibited the ultra-structure damage in rats after MCAO;riboflavin protec-ted the viability ( P <0.01 ) and the ultrastructure of neurons exposed to OGD .The enzyme activity of an-tioxidant SOD1 ( P <0.01 ) , CAT ( P <0.01 ) and GSH-Px ( P <0.01 ) was protected by riboflavin in MCAO model .No difference was found in the activity of SOD2 . Conclusion Riboflavin inhibits ischemia brain damage , and the protection of the activity of an-tioxidants is involved in the mechanism .

ABSTRACT

Effect of different water conditions on the physiological indexes (e.g.seed water content, vigor, antioxidase activities)of Panax notoginseng seeds were studied under process of after-ripening and germination.The results showed show that compared with 2.5% treatment, under the treatment of 5%, P.notoginseng seeds possessed stable seed water content, the seed vigor was exceed by 51%,variation of antioxidant enzyme (SOD, POD, CAT) activity and malondialdehyde (MDA) content were small, crude fat and total sugar content decreased significantly.With the increase of PEG 6000 concentration, the germination characteristic indexes obviously decreased, antioxidase activities increased firstly and decreased afterwards, content of MDA, soluble protein and total sugar increased obviously.There were significant positive correlation between germination characteristic indexes and osmotic substance content(r>0.900, P<0.01), and significant negative correlation with MDA (r>0.900, P<0.01).In conclusion, because the characteristic of dehydration intolerance of P.notoginseng seeds, 5% water content of sand burying stratification treatment was the best for after-ripening, 15% concentration of PEG 6000 treatment was the highest tolerance limit of germination process.

ABSTRACT

Objective: To study the effect of different concentration of plumbum (Pb) stress on antioxidase activity in different organs of Rabdosia Rubescentis Folium (RRF, the aerial parts of Rabdosia rubescens) and their growth, and provide the basis for the growth regulation of R. rubescens under Pb pollution. Methods: Ramets hydroponic test was performed with the concentration of Pb (0, 135, 270, and 540 mg/L) and the stress time (50 d), taking superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), relative growth rate (RGR), biomass, and Pb mass fraction as indexes, the determination of antioxidant enzyme activity was performed with spectrophotometer, and plant's Pb mass fraction of plant with plasma emission spectrometer was carried out. Results: Under Pb stress at different concentration (135-540 mg/L), SOD activity in the leaves and the roots of R. rubescens was increased first and then decreased, but no significant difference on SOD activity in the leaves at each level; POD activity in the leaves under Pb concentration (135-270 mg/L) was no significant difference compared with the control group, but was significantly increased at the concentration of 540 mg/L, while the POD activity in the roots under Pb stress at various concentration was significantly lower than that of the control group which gradually went up after declined; CAT activity in the leaves increased significantly with Pb concentration rising, and the activity in the root decreased gradually with Pb concentration rising. No significant effect on the growth of R. rubescens at low and mid levels of Pb concentration (135 and 270 mg/L), but under high concentration (540 mg/L) of Pb treatment compared with the control group, the biomass and RGR decreased by 33.33% and 46.59%, respectively. Conclusion: The stimulus response is different obviously for antioxidase activities between the leaves and roots of R. rubescens; Through fitting model predictive analysis, the critical Pb concentration is 301.99 mg/L, which decreases RGR by 10% compared with control group (P < 0.01). RRF shows a strong resistance to Pb stress.

ABSTRACT

Objective: To investgate the effects of NaCl stress on the growth and physiological characteristics of Thermopsis laceolata callus, and to reveal the physiological mechanism of T. laceolata under salty environment at the cellular level. Methods: The callus was cultured on the subculture medium with NaCl (0.2%-1.2%), and the contents of soluble sugar, soluble proteins, proline, and malondialdehyde (MDA) and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were determined and analyzed. Results: The callus growth expressed a decreasing trend with NaCl concentration increasing (≤ 0.8%) but had no significant difference to contrast. Nevertheless, the callus grew very slowly with NaCl concentration increasing (> 0.8%); the contents of soluble sugar and free proline increased firstly and then decreased, but were higher than those of the control group; the content of soluble protein and the activities of SOD, CAT, and POD increased firstly and then decreased; MDA accumulated slowly at salinity (≤ 0.8% NaCl), then increased rapidly at salinity (> 0.8% NaCl). Conclusion: T. laceolata callus has an adaptability to salty environment (≤ 0.8% NaCl), and the callus could alleviate the salty injury by increasing the contents of the osmotic regulation substances, such as proline and soluble sugar, decreasing the osmotic potential, and increasing the activities of anti-oxidative enzyme.

ABSTRACT

Objective: In order to get the method of improving the salt resistance of seeds and seedlings for Perilla frutescens under NaCl stress, seed germination and physiological characteristics of P. frutescens seedlings were studied. Methods: Several physiological indexes of P. frutescens seeds treated by 5-aminolevulinic acid (5-ALA) were measured. And other indexes of the seedlings like relative water content (RWC), the biomass, the content of soluble sugar, and malondialdehyde (MDA), the activities of superoxide (SOD), peroxidase (POD), and catalase (CAT) were also measured. Results: The germination indexes of P. frutescens seeds under NaCl stress have a obvious inhibition. But after the treatment at different concentrations of 5-ALA every germination index were all increased. And the seeds treated by 5-ALA with the concentration of 50 mg/L have the most significantly increase in every index. The germination vigor was 71.3%, the germination rate was 90.5%, the germination index and vigor index were 15.9 and 0.129 6, respectively. Every treatment of them coule improve the biomass and soluble sugar of the seedlings; The RWC of P. frutescens leaves decreased under NaCl stress, but after treated by 5-ALA, the rate of decrease has been relieved and the content of MDA in leaves was decreased. The activities of three enzymes including SOD, POD, and CAT were all increased. And the treatment of 5-ALA with the concentration of 50 mg/L got the maximin with 0.79, 7.69, and 5.84 U/mg, respectively. Conclusion: 5-ALA with the appropriate concentration of 50 mg/L could significantly alleviate the damages to the seeds and seedlings of P. frutescens under NaCl stress and promote the salt resistance of the seeds and seedlings.

ABSTRACT

Objective To compare the changes in nitric oxide (NO), total ability of anti-oxidize antioxidase, superoxide dismutase (SOD), glutathion peroxidase (GSH-PX), catalase (CAT) and maleic dialdehyde(MDA) in rats after single and accumulative 60Co γ-irradiation . Methods 48 rats were randomly divided into two groups of single and accumulative irradiation. Each group was irradiated by 60Co gamma-rays. The total irradiation doses were 0.4, 0.8, 1.6, 3.2 and 6.4 Gy, respectively in each group. The NO, total ability of anti-oxidize antioxidaze, SOD, GSH-PX, CAT and MDA in serum were measured at day 1 after last irradiation. Results Compared with the single irradiation group, the NO (52.6-117.9 μmol/ml), total ability of anti-oxidize antioxidaze (3.3-26.2 U/ml), the antioxidase activity of the SOD (26.3-167.5 U/ml), GSH-PX (740.8-2462.4 U/ml), CAT (3.3-29.4 U/ml) and the content of MDA(29.3-155.1 nmol/ml) of mt serum in accumulative irradiation group were increased after irradiation, which was related with the accumulative irradiation dose. For instance, total ability of anti-oxidize antioxidase (26.2 U/ml), antioxidase activity of the SOD (167.5 U/ml) and CAT (29.4 U/ml) in 0.4 Gy group of accumulative irradiation were significandy increased when compared with those of control group. However, the content of MDA in accumulative irradiation group was obviously higher than that in single radiation group when the irradiation doses delivered over 3.2 Gy, which might be correlated with higher antioxidase activity. Conclusions Low dose of accumulative gamma-rays irradiation can induce the stimulative effect of antioxidase activity. However, higher dose of accumulative gamma-rays irradiation can damage the activity of antioxidase.

ABSTRACT

AIM:To investigate protective effect of ethanol extract of Forsythia suspensa flower(FFE)on mitochondrial injury induced by free radicals,antioxidation and superoxide anion(O ?-__2)scavenging activities and anti-aging mechanism.METHODS:Thiobarbituric acid(TBA)colorimetric method was used to determine malondialdehyde(MDA).Spectrometric method was used to measure the swelling of mitochondria and activities of scavenging O ?-__2 which was produced by reduced nicotinamide adenine dinucleotide(NADH)-4-Nitrobluetetrazolium chloride(NBT)-N-Methylphenazonium methyl sulfate(PMS)system.The neck back of mice were injected subcutaneously with D-galactose to induce aging model at a dose of 100 mg/(kg?d)for 7 weeks.The effects of FFE on the activities of catlase(CAT),surperoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)and anti-hydroxyl radical were assayed by ammonium molybdate colorimetry,xanthine oxidase method,dithiodinitrobenzoic acid colorimetry and Fenton reaction colorimetry in vivo respectively.RESULTS:FFE could inhibit mitochondrial injury and swelling induced by Fe 2+-L-Cys in a concentration-dependent manner and also had significant O ?-__2 scavenging effect.Moreover,the activities of CAT,SOD,GSH-Px and anti-hydroxyl radical in mice liver homogenate were increased significantly by FFE.CONCLUSION:FFE protects mitochondria by scavenging reactive oxygen species,and increasing the activities of antioxidase.So FFE has the pharmaceutical values of antioxidant and anti-aging.
